Position Focus:

Staff Scientist position available in the FIB-SEM Collaboration Core (F-SCC) of Yale School of Medicine. The F-SCC is a central core facility using the state-of-the-art enhanced Focused Ion Beam Scanning Electron Microscopy (FIB-SEM) technology for large volume imaging with nanometer isotropic resolution. Under supervision of the Directors of F-SCC, the candidate will go through comprehensive training by the leading experts in the field to be familiar with the entire sample preparation and imaging pipeline including: electron microscopy (EM) sample preparation of biological specimens, FIB-SEM sample preparation guided by x-ray micro-CT, enhanced FIB-SEM imaging, post data processing, and image analysis using deep learning neural network approach. After training, the candidate will primarily focus on automating EM sample preparation pipeline to enable best possible ultrastructure preservation and heavy metal staining contrast of biological samples. The candidate is expected to work closely with the internal and external collaborators and group members.

While the job duty may evolve according to the needs of the organization and the interests of the individual, specific responsibilities can include: Develop automated EM sample preparation pipeline with focus on clinical applications. Prepare biological specimens (tissues, cultured cells, etc.) using heavy metal staining followed by resin embedding. Evaluate sample quality using transmission electron microscopy (TEM). Prepare sample for FIB-SEM imaging: mount and trim samples to target specific region of interest using ultramicrotome guided by x-ray micro-CT. Perform three-dimensional volume EM image acquisition using the enhanced FIB-SEM platform. Perform alignment of raw image data using image registration pipeline to form isotropic 3D image datasets. Perform image analysis using deep learning neural network image segmentation software packages.

1. Designs, plans, implements, and evaluates complex multifaceted research projects/systems, including method development, adaptation, and validation. 2. Collaborates with internal and external researchers to define research endeavors and the development of research hypothesis and approach, discuss and evaluate research results, and to ensure the smooth and safe operation of the laboratory. 3. Solves complex methodology, protocol, procedural and research problems through design of techniques, procedures and policies that will achieve research goals. 4. Develops long range plans for supplies and equipment to ensure smooth operation of laboratory. 5. Investigates, analyzes, and evaluates complex data, data collection systems and methods to reach scientific conclusions and ensures the integrity of research data. 6. Collaborates with supervisors to coordinate major lab renovations working with Facilities, Health and Safety, and Purchasing to ensure deadlines, safety standards and purchases are according to plans and within budgets. 7. Prepares scientific reports and papers for research proposals and published reports. 8. May perform other duties as assigned.

Essential Duties

1. Manages the daily activities of a research resource laboratory. Oversees the scheduling, use, and availability of instrumentation, supplies, and specialized work areas. 2. Carries out biotechnology research design, develop, and modify procedures and instrument operating programs. 3. Develops, interprets, and analyzes the results of biotechnological analyses and/or syntheses. 4. Investigates, analyzes, and evaluates alternative instrumentation, methodologies and techniques to maintain a state-of-the-art biotechnology resource laboratory. 5. Recommends and develops new protocols and initiates and adapts new techniques to resolve problems. 6. Conducts research to determine the feasibility and scope of providing new/revised services. Develops and evaluates new techniques and equipment. 7. Prepares budgets and reports pertaining to laboratory costs and income. 8. Determines and establishes policies and procedures on laboratory safety consistent with University safety regulations. 9. May perform other duties as assigned.

Required Education and Experience

Master’s Degree in chemistry, biology, biochemistry, or other related discipline and three years of experience or an equivalent combination of experience and education.

Required Skill/Ability 1:

Ability to learn and develop new heavy-metal staining protocols to support the advances in volume electron microscopy.

Required Skill/Ability 2:

Proficiency in using diverse software tools. Ability to learn and operate instrument and troubleshoot associated issues.

Required Skill/Ability 3:

Ability to participate in writing of grant proposals and manuscripts. Ability to anticipate project needs and plan carefully to optimize resources necessary for successful project completion.

Required Skill/Ability 4:

Ability to search and interpret scientific papers, protocols, standard operating procedures, safety standards, maintenance instructions and procedure manuals for laboratory equipment.

Required Skill/Ability 5:

Ability to maintain laboratory facilities and work areas with the flexibility to learn and adapt to new procedures incidental to laboratory activities.

Preferred Education, Experience and Skills:

PhD Degree plus three years of experience or Master’s Degree plus six years of experience, or an equivalent combination of experience and education. Degree in chemistry, biology, engineering, biochemistry, or other related discipline preferred. Special considerations will be given to candidates who have hands-on experience and strong interest in EM sample preparation and automation.
